  • (i) Fix (preserve) bean root tips in acetic alcohol for 24 hours.

    (ii) Soak the tips in HCl. This helps to soften the cells and allows the stain to be absorbed easier.

    (iii) Rinse with distilled water.

    (iv) Soak the tips in Feulgens solution to stain the DNA.

    (v) Transfer to a microscope slide.

    (vi) Add acetic orcein to intensify the stain.

    (vii) Apply a coverslip and cover with filter paper. Squash to spread out the tissue. Soak up the excess stain with the filter paper.

    (viii) Examine under low and high power.
